MUC1 100mer peptide is a synthetic cancer vaccine composed of a 100-amino acid sequence derived from the variable number tandem repeat (VNTR) region of the mucin-1 (MUC1) antigen. This region consists of five tandem repeats, each containing twenty amino acids. The vaccine is designed to induce immune responses against tumor cells that overexpress and aberrantly glycosylate the MUC1 protein—a common feature in various cancers such as pancreatic, colon, bile duct, and lung cancers. The non-glycosylated form used in clinical trials exposes immunogenic epitopes not present on normal epithelial cells. When administered with adjuvants like SB-AS2 or Poly-ICLC, it has demonstrated safety and modest immunogenicity by eliciting both humoral (antibody-mediated) and cellular (T-cell mediated) anti-MUC1 responses in early-phase clinical studies[2][3][6][9]. Its primary mechanism is to stimulate an adaptive immune response targeting tumor-associated forms of the MUC1 antigen.
