MUC1 antibody-functionalized microparticles are an experimental diagnostic and targeted delivery system designed to detect and visualize colorectal cancer (CRC) lesions. These microparticles utilize a "universal" surface functionalization approach, often employing a Protein L scaffold (APL-MPs) to incorporate antibodies specific to Mucin-1 (MUC1). MUC1 is a heavily glycosylated transmembrane protein that is significantly overexpressed on the surface of various epithelial cancers, including colorectal, breast, and pancreatic cancers. By binding specifically to MUC1 antigens on the luminal surface of the colon, these microparticles facilitate the detection of heterogeneous tumor lesions that might be missed by conventional visualization techniques. Preclinical evaluations in 3D tumoroids and orthotopic animal models have demonstrated that these particles can identify MUC1-positive tissues in proportion to antigen expression, serving as a modular platform for precision imaging of diverse tumor phenotypes.
