Overview

MUC1-aptamer S2.2 is a 25-nucleotide single-stranded DNA (ssDNA) aptamer that selectively binds to the Mucin 1 (MUC1) glycoprotein with high affinity (Kd = 0.135 nM). It specifically targets the APDTRPAPG sequence within the variable number tandem repeat (VNTR) region of MUC1, a protein that is frequently overexpressed and aberrantly glycosylated in various adenocarcinomas. The aptamer's unique three-dimensional structure allows it to distinguish MUC1-expressing cancer cells, such as A-549 lung cancer cells, from normal tissues. In research settings, S2.2 is primarily utilized as a targeting moiety to deliver cytotoxic agents like doxorubicin or imaging contrast agents directly to tumor sites, aiming to improve therapeutic indices and diagnostic precision.
