Overview

MUC1 CAR-T is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed to target Mucin 1 (MUC1), a transmembrane glycoprotein that is frequently overexpressed and aberrantly glycosylated in various epithelial malignancies. Developed by Henan Province Cancer Hospital, this cell therapy involves the genetic modification of a patient's own T cells to express a CAR construct specific for the MUC1 antigen, typically incorporating costimulatory domains such as 4-1BB to enhance T-cell persistence and anti-tumor activity. MUC1 plays a critical role in cell signaling, lubrication, and protection of healthy epithelium, but its overexpression in pancreatic cancer and other solid tumors promotes tumor cell survival, invasion, and resistance to chemotherapy. By specifically recognizing MUC1-positive cells, the CAR-T cells initiate a targeted cytotoxic response aimed at eradicating the tumor while potentially overcoming the immunosuppressive microenvironment of solid tumors.
