The MUC1 glycopeptide vaccine is an investigational therapeutic and preventive cancer vaccine designed to induce immune responses against the mucin 1 (MUC1) protein, a transmembrane glycoprotein that is overexpressed and aberrantly glycosylated in many epithelial cancers, including breast, colon, ovarian, and pancreatic cancers. The vaccine typically consists of synthetic peptides or glycopeptides derived from the tandem repeat region of MUC1 containing tumor-associated carbohydrate antigens (TACAs), such as Tn antigen (GalNAc-α-O-Ser/Thr). These constructs are often conjugated to immunogenic carriers or adjuvants to enhance uptake by dendritic cells via receptors like macrophage galactose-type lectin (MGL), promoting antigen presentation and robust humoral and cellular immune responses. Clinical studies have shown that vaccination can elicit anti-MUC1 IgG antibodies in a subset of recipients; responders may experience reduced recurrence rates of precancerous lesions such as colon adenomas. The approach aims to break tolerance against self-antigen by targeting tumor-specific post-translational modifications[6][7][9].
