Drug intelligence / Profile preview
MUC1-KLH + QS-21 is an investigational therapeutic cancer vaccine developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of high-risk breast cancer. The vaccine is composed of a synthetic 30-amino acid peptide representing 1.5 repeats of the MUC1 (mucin-1) extracellular tandem repeat sequence, which is chemically conjugated to the carrier protein keyhole limpet hemocyanin (KLH) using an MBS bifunctional linker. This conjugate is administered in combination with the saponin-based immunological adjuvant QS-21. The vaccine targets the MUC1 glycoprotein, which is frequently overexpressed and underglycosylated on the surface of breast cancer cells. Its primary mechanism of action is to stimulate the patient's immune system to produce high-titer IgM and IgG antibodies against MUC1-expressing tumor cells. The vaccine is typically evaluated in the adjuvant setting for patients who are free of detectable disease after standard primary therapy but remain at high risk for recurrence.
