MUC1-mRNA dendritic cells (MUC1-DCs) is an autologous cell-based immunotherapy designed to treat cancers overexpressing the Mucin 1 (MUC1) glycoprotein, such as pancreatic adenocarcinoma. The therapy involves harvesting patient-derived monocytes, culturing them with GM-CSF and IL-4 to generate immature dendritic cells, and maturing them with TNF-α. These mature dendritic cells are then transfected with mRNA encoding the MUC1 antigen via electroporation. Once administered intradermally, these DCs present MUC1 epitopes to the patient's immune system, stimulating a targeted T-cell response against MUC1-positive tumor cells. Clinical studies have primarily evaluated this vaccine in combination with MUC1-specific cytotoxic T lymphocytes and gemcitabine for unresectable or recurrent pancreatic cancer.
