Overview

The MUC1 peptide + poly ICLC vaccine is an investigational cancer immunoprevention vaccine designed to prevent the recurrence of advanced colorectal adenomas and their progression to colorectal cancer. Developed by researchers Robert E. Schoen and Olivera J. Finn at the University of Pittsburgh, the vaccine targets the tumor-associated antigen MUC1, which is abnormally glycosylated and overexpressed in adenomatous polyps and colorectal cancer. The formulation consists of a 100-amino acid synthetic MUC1 peptide (100 µg) combined with Poly ICLC (Hiltonol), a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) agonist to enhance the immune response. By training the immune system to recognize and eliminate cells expressing the abnormal MUC1 protein, the vaccine aims to provide long-term protection against colorectal malignancy. Clinical trials have demonstrated that immune responders to the vaccine show a significant reduction in adenoma recurrence compared to placebo.
