Overview

The MUC1 peptide-poly-ICLC vaccine is a subunit cancer immunotherapy composed of a synthetic 100-amino acid segment of the mucin 1 (MUC1) protein combined with the adjuvant polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose (poly-ICLC, also known as Hiltonol). The vaccine is designed to stimulate an immune response against tumor cells expressing aberrant or overexpressed forms of the glycoprotein antigen MUC1. Poly-ICLC acts as a Toll-like receptor 3 (TLR3) agonist to enhance immunogenicity by promoting dendritic cell activation and cytotoxic T-cell responses. The primary indications under investigation include prevention of recurrence in patients with advanced adenomas (precancerous colon polyps), treatment or prevention in high-risk populations for lung cancer, and as an adjunct therapy in breast ductal carcinoma. Clinical trials have demonstrated that the vaccine can induce anti-MUC1 IgG responses in a subset of recipients; immune responders may experience reduced rates of adenoma recurrence compared to nonresponders[5][6][7][9].
