Overview

The MUC1 peptide vaccine is a therapeutic cancer vaccine developed by Dr. Olivera Finn at the University of Pittsburgh. It consists of a synthetic peptide derived from the tandem repeat domain of the Mucin 1 (MUC1) protein, a transmembrane glycoprotein that is frequently overexpressed and abnormally hypoglycosylated in various adenocarcinomas. The vaccine is designed to induce a cellular immune response, specifically activating CD8+ and CD4+ T cells to recognize and eliminate cells expressing the tumor-associated form of MUC1 (tMUC1). In clinical settings, it is typically administered in combination with the adjuvant Hiltonol (poly-ICLC), a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) agonist to enhance the immunogenicity of the peptide. The vaccine is being investigated for immune interception and prevention in conditions such as ductal carcinoma in situ (DCIS) and colorectal adenomatous polyps.
