MUC13-PPNPs is an antibody-targeted nanoparticle formulation designed for the treatment of pancreatic ductal adenocarcinoma (PDAC). The drug consists of paclitaxel (PTX) encapsulated within multi-layered poly(lactic-co-glycolic acid) (PLGA) nanoparticles (PPNPs) that are stabilized with Pluronic F127 and polyvinyl alcohol, and coated with poly-L-lysine. The nanoparticles are functionalized with an anti-MUC13 antibody to enable specific delivery to MUC13-overexpressing tumor cells. Beyond direct cytotoxicity, MUC13-PPNPs have been shown to influence the tumor microenvironment by repolarizing tumor-associated macrophages (TAMs) from an M2 to an M1 phenotype and downregulating oncogenic signaling pathways such as sonic hedgehog. It is currently being investigated in preclinical models, where it has demonstrated synergy with gemcitabine.
