Overview

MUC16-CAR T cells are **chimeric antigen receptor (CAR) T cell therapies genetically engineered to recognize and target the MUC16 (also known as CA-125) antigen**, which is overexpressed on the surface of most ovarian cancers and certain other gynecologic malignancies. These therapies use T cells modified to express a synthetic receptor (typically derived from the antibody 4H11) that binds specifically to the extracellular domain of MUC16, thereby directing T-cell-mediated cytotoxicity towards MUC16-positive tumor cells. In advanced research, some MUC16-CAR T cells have been 'armored' (e.g., to secrete an anti-WT1 BiTE or interleukin-12) to enhance activity and overcome tumor heterogeneity and immune suppression within the tumor microenvironment[1][4][5]. Early-phase clinical trials have shown safety and some evidence of antitumor activity in patients with recurrent or refractory epithelial ovarian, peritoneal, or fallopian tube cancers[1][5][7]. The primary indications are advanced ovarian cancer and related high-MUC16-expressing solid tumors.
