Overview

This investigational autologous CAR T-cell therapy, developed by the Shenzhen Geno-Immune Medical Institute (GIMI), is designed for the treatment of relapsed and refractory multiple myeloma. Utilizing the proprietary 4SCAR2.0 (fourth-generation safety-designed CAR) platform, the therapy involves the administration of T cells engineered to target multiple tumor-associated antigens, including B-cell maturation antigen (BCMA), CD38, CD138, and GPRC5D. This multi-target approach aims to mitigate tumor antigen escape, a common cause of relapse in single-target CAR T therapies. The 4SCAR2.0 construct incorporates a CD27 costimulatory domain to enhance cytokine signaling and immune memory, along with an inducible Caspase-9 safety mechanism that allows for the rapid elimination of CAR T cells in the event of severe toxicity, such as cytokine release syndrome (CRS).
