Overview

multi-rBCG is a recombinant DNA vaccine derived from Bacille Calmette-Guérin (BCG), an attenuated strain of Mycobacterium bovis. It is being developed as an immunotherapy, primarily for the treatment of bladder cancer. This genetically engineered version of BCG aims to enhance its inherent immunomodulatory activities, for instance, by expressing specific antigens, cytokines, or other immune-modulatory molecules, or by overexpressing small molecules like STING agonists. The therapeutic goal is to improve antitumor efficacy and address limitations associated with wild-type BCG, such as variable response rates and potential side effects. Its mechanism involves stimulating both innate and adaptive immune responses, leading to the recruitment of various immune cells to the tumor microenvironment and exerting direct cytotoxic effects on cancer cells.
