Overview

The **multiepitope autologous dendritic cell vaccine** is an investigational cellular immunotherapy developed primarily at the University of North Carolina at Chapel Hill. It consists of a patient's own (autologous) dendritic cells that are harvested via apheresis, expanded in vitro, and pulsed with multiple human epidermal growth factor receptor 2 (HER2/neu)-derived peptides, specifically the **E75** (nelipepimut-S) and **E90** epitopes. These dendritic cells function as professional antigen-presenting cells that, when re-administered to the patient, present the HER2 epitopes to the immune system to stimulate a robust cytotoxic T-lymphocyte (CTL) response against HER2-expressing tumor cells. This vaccine has been studied in Phase II clinical trials for locally recurrent or metastatic breast cancer, often administered in combination with trastuzumab and chemotherapy (such as vinorelbine) to enhance therapeutic efficacy.
