MURA-7012

MURA-7012 is an engineered interleukin-12 (IL-12) immunotherapy developed by Mural Oncology for the treatment of cancer. The drug is designed to leverage the potent anti-tumor activity of native IL-12 while mitigating its hallmark systemic toxicity. This is achieved by splitting the IL-12p70 heterodimer into two inactive monomers (IL12p35 and IL12p40), which are separately fused to antibody fragments and sequentially injected. These subunits preferentially self-assemble at the tumor site, concentrating active IL-12 in the tumor microenvironment and limiting systemic exposure. Preclinical studies have shown that this approach reduces serum levels while maintaining effective concentrations within tumors, potentially reducing toxicities associated with systemic administration[1][3][4][5][7][8].
