MURAL-002 is an engineered, decoy-resistant interleukin-18 (IL-18) cytokine candidate developed by Mural Oncology. Wild-type IL-18 is a potent pro-inflammatory cytokine that stimulates the production of interferon-gamma (IFN-γ) and activates T cells and natural killer (NK) cells; however, its therapeutic utility is limited by high levels of IL-18 binding protein (IL-18BP) in the tumor microenvironment, which acts as a decoy receptor and neutralizes IL-18. MURAL-002 is designed to maintain high affinity for the IL-18 receptor while being resistant to neutralization by IL-18BP. This allows the drug to effectively stimulate anti-tumor immune responses in advanced solid tumors, including platinum-resistant ovarian cancer, where endogenous IL-18 activity is typically suppressed.
