Murine anti-PD-1 antibody is a monoclonal antibody used in preclinical research to block the Programmed Cell Death Protein 1 (PD-1) receptor in mouse models. PD-1 is an immune checkpoint receptor expressed on T cells that, upon binding to its ligands PD-L1 or PD-L2, inhibits T-cell activity to maintain peripheral tolerance and prevent autoimmunity. In syngeneic cancer models, such as those for pancreatic, hepatocellular, breast, and lung cancers, murine anti-PD-1 antibodies are employed to study the effects of checkpoint blockade on the tumor microenvironment. These antibodies promote anti-tumor immunity by increasing the infiltration of effector cells like CD8+ T cells and NK cells while decreasing immunosuppressive populations such as regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs). They are frequently evaluated in combination with other immunotherapies, such as TLR3 agonists, to enhance therapeutic efficacy.
