Murine antithymocyte globulin (mATG) is a polyclonal rabbit-derived antibody directed against mouse thymocytes, serving as a murine surrogate for the clinical immunosuppressant Thymoglobulin (rabbit anti-human thymocyte globulin). It is primarily used in preclinical research to study T-cell depletion and immune modulation in models of solid organ transplantation, hematopoietic stem-cell transplantation, and autoimmune diseases like type 1 diabetes. The drug's mechanism involves the depletion of various T-cell subsets, predominantly mediated by macrophages in vivo, though it also utilizes the Fas/FasL pathway to target regulatory T cells. Beyond simple depletion, mATG has been shown to induce immunoregulation and alter disease progression by enhancing the functional activity of regulatory T cells and promoting homeostatic expansion of naive T cells.
