Overview

murine BCMA-CART (also known as ARI2m) is a chimeric antigen receptor T-cell (CAR-T) therapy engineered to target B-cell maturation antigen (BCMA), which is highly expressed on malignant plasma cells in multiple myeloma. The extracellular antigen-recognition domain of this CAR is derived from a murine (mouse) single-chain variable fragment specific for human BCMA. Upon infusion into the patient, these genetically modified autologous T cells recognize and kill BCMA-expressing tumor cells through cytotoxic immune mechanisms. Preclinical studies demonstrated efficacy against multiple myeloma; however, the use of a non-human (murine) scFv can lead to immunogenicity and potential immune rejection by patients[1][3]. This has led to the development of humanized versions with improved safety and efficacy profiles.
