Overview

Murine interleukin-11 (mIL-11) is a cytokine belonging to the gp130 family. It exerts pleiotropic actions on various cell types, including hematopoietic, hepatic, stromal, epithelial, neural, and osteoclast cells. The mechanism of action involves the formation of a high-affinity receptor complex with the interleukin-11 receptor (IL-11R) and the transmembrane signaling receptor gp130, which subsequently induces gp130-dependent signaling. Structural and functional studies have identified three distinct receptor binding sites on mIL-11, analogous to those found in interleukin-6, supporting the model of a hexameric receptor complex. Murine IL-11 has been shown to stimulate proliferation in murine plasmacytoma cells, primitive bone marrow progenitor cells, megakaryocyte precursors, and hippocampus neuronal progenitor cells. Its expression is widespread in normal murine tissues, with notable levels in the hippocampus and developmentally regulated expression in the testis, suggesting roles in neural and testicular function, including accelerating spermatogenesis recovery after cytotoxic therapy.
