Overview

Murine PVSRIPO is a mouse-adapted version of PVSRIPO, a recombinant oncolytic polio-rhinovirus chimera derived from the Sabin type 1 attenuated poliovirus. It was engineered to overcome the natural resistance of murine cells to poliovirus infection by adapting the virus for competence in mouse models, specifically by ensuring functionality of the internal ribosome entry site (IRES) and receptor binding in murine tissues. The virus targets CD155 (the poliovirus receptor), which is widely expressed on various tumor cells and within the tumor microenvironment. Upon infection, murine PVSRIPO induces immunogenic cell death and activates innate and adaptive immune responses, making it a critical tool for studying oncolytic virotherapy in syngeneic immunocompetent mouse models of glioblastoma and melanoma.
