The murine-specific anti-DLL4 monoclonal antibody is a research-grade biologic developed to specifically target and inhibit the Delta-like ligand 4 (DLL4) in mice. DLL4 is a critical component of the Notch signaling pathway, playing a vital role in tumor angiogenesis and the maintenance of cancer stem cells (CSCs). In preclinical oncology research, particularly studies conducted by OncoMed, this antibody serves as a surrogate to evaluate the effects of blocking host-derived (murine) DLL4, as opposed to tumor-derived (human) DLL4 in xenograft models. Inhibition of murine DLL4 leads to "unproductive angiogenesis," characterized by an increase in non-functional vascular sprouting and branching, which results in poor tumor perfusion and inhibited growth. This agent is often used in combination with human-specific anti-DLL4 antibodies (such as demcizumab) to achieve a dual blockade of both the tumor and the host stroma, providing insights into the therapeutic potential of targeting the Notch pathway in various cancers, including pancreatic cancer.
