Overview

Murine surrogate BCMA-TCE is a preclinical bispecific T-cell engager (TCE) developed by Pfizer for use in syngeneic mouse models, such as the Vk*MYC multiple myeloma model. It is designed to mimic the activity of human BCMA-targeted TCEs (e.g., elranatamab) by simultaneously binding to murine B-cell maturation antigen (BCMA) on malignant plasma cells and the CD3 epsilon subunit on murine T cells. This dual binding facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent lysis of the tumor cells. In research settings, it is utilized to investigate dosing schedules and mechanisms of acquired resistance, including genomic antigen escape via retrotransposition of intracisternal A-particle (IAP) elements into the Tnfrsf17 gene and transient downregulation of BCMA expression.
