muSTAT3 ASO is a generation 2.5 antisense oligonucleotide (ASO) designed to target and inhibit the expression of murine Signal Transducer and Activator of Transcription 3 (STAT3) mRNA. It serves as a murine-specific surrogate for the clinical candidate AZD9150 (danvatirsen), which targets human STAT3. Developed by AstraZeneca, muSTAT3 ASO is primarily used in preclinical research to investigate the pharmacodynamics of STAT3 inhibition within the tumor microenvironment (TME). Treatment with this ASO has been shown to significantly reduce immunosuppressive cell populations, including M2-polarized macrophages (CD163+) and myeloid-derived suppressor cells (MDSCs), thereby enhancing the antitumor activity of T-cell targeted therapies such as anti-PD-L1 antibodies in various syngeneic and genetically engineered mouse models.
