Mutacin 1140 is a class I lantibiotic produced by the bacterium *Streptococcus mutans* JH1140. It is a ribosomally synthesized and post-translationally modified peptide (RiPP) antibiotic that exhibits potent activity against Gram-positive bacteria, including methicillin-resistant *Staphylococcus aureus* (MRSA), vancomycin-resistant enterococci (VRE), *Clostridium difficile*, and *Streptococcus pneumoniae*. The mechanism of action of mutacin 1140 is two-fold: it binds to the cell wall precursor lipid II to inhibit bacterial cell wall synthesis and also disrupts bacterial membranes by forming a complex around its target. Unlike nisin, another class I lantibiotic with which it shares structural similarities in rings A and B, mutacin 1140 does not form membrane pores but still effectively kills bacteria. Mutacin 1140 analogs have been engineered for improved bioactivity and pharmacokinetic properties. The drug is under preclinical development as a novel therapeutic for serious Gram-positive infections such as MRSA bacteremia and *C. difficile*-associated diarrhea.
