Overview

Mutant KRAS G12V-specific TCR transduced autologous T cells is a cell therapy that uses a patient's own T cells genetically engineered to express a T cell receptor (TCR) that specifically recognizes the KRAS G12V mutation presented by HLA-A*11:01. This therapy targets cancer cells harboring the KRAS G12V mutation, which is a common driver mutation in various solid tumors, particularly pancreatic, colorectal, and non-small cell lung cancers. The theoretical basis is that these engineered T cells will target and kill HLA-matched mutant KRAS cancer cells while sparing normal cells. Some versions of this therapy (like AFNT-211) include additional engineering features such as the wildtype CD8α/β coreceptor and a FAS-41BB switch receptor to enhance anti-tumor activity and overcome immunosuppression in the tumor microenvironment.
