Mutant KRAS TCR-T therapy is an autologous adoptive cell therapy developed by Anocca AB that utilizes engineered T cells to target specific mutations in the KRAS gene, such as G12D and G12V. The therapy involves the ex vivo modification of a patient's T cells to express high-affinity T-cell receptors (TCRs) identified through Anocca's proprietary precision targeting platform. These TCRs are designed to recognize neoantigenic peptides derived from mutant KRAS proteins when presented by specific Human Leukocyte Antigen (HLA) alleles on the surface of tumor cells. This approach is particularly relevant for pancreatic ductal adenocarcinoma (PDAC), where KRAS mutations are nearly ubiquitous, as well as colorectal and non-small cell lung cancers. By targeting intracellular driver mutations via the MHC/HLA pathway, TCR-T therapy overcomes the limitations of CAR-T cells, which are restricted to surface-expressed antigens.
