Overview

Muvalaplin (LY3473329) is an orally active, selective small molecule inhibitor of lipoprotein(a) [Lp(a)] formation. It works by disrupting the initial non-covalent interaction between apolipoprotein(a) [apo(a)] and apolipoprotein B100 [apoB100], thereby preventing the disulfide bond formation necessary for Lp(a) assembly. Elevated Lp(a) levels are associated with increased risk of cardiovascular diseases, and muvalaplin is being developed as a first-in-class oral therapy to lower Lp(a). The drug is under development by Eli Lilly, primarily targeting patients at high risk for cardiovascular events due to elevated Lp(a)[1][3][4][7][8].
