Drug intelligence / Profile preview
muzastotug + INCA33890 is an investigational combination immunotherapy consisting of muzastotug (ADG126), a masked anti-CTLA-4 monoclonal antibody (SAFEbody) developed by Adagene, and INCA33890, a bispecific antibody targeting TGFβR2 and PD-1 developed by Incyte (in collaboration with Merus). Muzastotug is designed to selectively deplete regulatory T cells (Tregs) in the tumor microenvironment by targeting a specific CTLA-4 epitope, while minimizing systemic toxicity through its masking technology. INCA33890 simultaneously blocks the PD-1 and TGF-β pathways, which are key drivers of immune evasion and T-cell suppression. The combination is being evaluated in a Phase 1 clinical trial sponsored by Incyte for the treatment of patients with third-line microsatellite-stable colorectal cancer (MSS CRC), with or without liver metastases.
