MV-m-uPA is a recombinant oncolytic measles virus (MV) engineered to specifically target the murine urokinase-type plasminogen activator receptor (uPAR). uPAR is a clinically relevant biomarker overexpressed in many aggressive cancers and is involved in tumor cell invasion and metastasis. The virus is retargeted by displaying a targeting ligand on the viral hemagglutinin (H) protein, which redirects viral entry from its natural receptors (such as CD46 or SLAM) to uPAR. Once inside the cell, the virus replicates, induces syncytia formation, and causes cell death (oncolysis), while also potentially stimulating an anti-tumor immune response. MV-m-uPA is primarily used in preclinical syngeneic mouse models to evaluate the efficacy and safety of uPAR-targeted virotherapy in an immunocompetent setting, serving as a murine-specific counterpart to the human-targeted version, MV-h-uPA.
