Overview

MV-PNP-anti-PSCA is an engineered oncolytic measles virus (MV) vaccine strain developed for the targeted treatment of pancreatic adenocarcinoma and potentially prostate cancer. The virus is genetically retargeted to enter cells exclusively via the prostate stem cell antigen (PSCA), which is highly expressed in pancreatic and prostate cancers but absent in normal tissues. Furthermore, the virus is 'armed' with the gene for purine nucleoside phosphorylase (PNP), a prodrug convertase. When administered alongside the prodrug fludarabine, PNP converts it into a highly toxic metabolite, inducing a potent bystander effect that kills both infected and neighboring uninfected tumor cells. Preclinical studies have demonstrated its efficacy in gemcitabine-resistant pancreatic cancer models.
