Overview

MVA-209-FSP-B1 is a viral vector-based cancer vaccine component developed by Nouscom, utilizing the Modified Vaccinia Ankara (MVA) platform. It is specifically designed as one of the 'boost' vectors in the NOUS-209 heterologous prime-boost vaccine regimen. NOUS-209 targets 209 shared frameshift peptide (FSP) neoantigens that are frequently generated in tumors with microsatellite instability-high (MSI-H) or mismatch repair deficiency (dMMR), such as colorectal, gastric, and endometrial cancers. Because of the large number of antigens encoded, the payload is distributed across multiple vectors (e.g., B1 and B2). The regimen typically begins with a priming dose using Great Ape Adenovirus vectors (GAd209-FSP) followed by boosting with MVA vectors like MVA-209-FSP-B1. The vaccine aims to induce a broad, polyfunctional CD8+ and CD4+ T-cell response against multiple tumor antigens simultaneously to overcome tumor heterogeneity and prevent immune escape.
