MVA-209-FSP-B4 is a viral vector-based cancer vaccine component developed by Nouscom. It is the Modified Vaccinia Ankara (MVA) boost component of the NOUS-209 genetic vaccine platform. NOUS-209 is designed to target 209 shared neoantigens, specifically frameshift peptides (FSPs), which result from mutations in microsatellite instability-high (MSI-H) and mismatch repair deficient (dMMR) tumors. The vaccine regimen typically involves a heterologous prime-boost strategy, where a Great Ape Adenovirus (GAd) vector (GAd20-209-FSP) is used for priming and MVA-209-FSP-B4 is used for boosting. This approach is intended to induce a robust, broad, and long-lasting CD8+ and CD4+ T-cell response against the tumor. It is currently being evaluated in clinical trials for MSI-H colorectal, gastric, and endometrial cancers, often in combination with checkpoint inhibitors like pembrolizumab.
