Overview

MVA-B is a recombinant modified vaccinia Ankara-based HIV-1 vaccine that simultaneously expresses monomeric gp120 as a cell-released product and Gag-Pol-Nef (GPN) as an intracellular polyprotein of 160 kDa. The gp120 Env protein comes from the HIV-1 primary isolate BX08, while the Gag-Pol-Nef components are derived from the IIIB HIV-1 isolate. The GPN polyprotein has been engineered by removing immunosuppressive sequences and preventing virus-like particle formation. As a therapeutic vaccine, MVA-B aims to enhance HIV-specific immune responses in individuals already infected with HIV-1. It works by stimulating both CD4+ and CD8+ T cell responses against HIV-1 antigens, inducing new responses, expanding pre-existing ones, and generating polyfunctional T cell responses.
