MVA-BN-Brachyury is a therapeutic cancer vaccine developed by Bavarian Nordic in collaboration with the National Cancer Institute (NCI). It utilizes the Modified Vaccinia Ankara (MVA) viral vector, which is non-replicating in human cells, to deliver the gene for Brachyury, a transcription factor overexpressed in various solid tumors (such as chordoma and metastatic carcinomas) but rarely in normal adult tissues. Brachyury is associated with the epithelial-mesenchymal transition (EMT), which promotes tumor metastasis and resistance to therapy. The vaccine often incorporates the TRICOM (TRIad of COstimulatory Molecules) complex, consisting of B7-1, ICAM-1, and LFA-3, to enhance the activation of T cells against Brachyury-expressing cancer cells. It is being investigated as a monotherapy and in combination with other treatments like radiation or booster vaccines (e.g., FPV-Brachyury).
