Overview

MVA-BN-HIV is an investigational recombinant viral vector vaccine developed by Bavarian Nordic in collaboration with Janssen (Johnson & Johnson) and the Beth Israel Deaconess Medical Center. It utilizes the Modified Vaccinia Ankara (MVA) platform—a highly attenuated, non-replicating orthopoxvirus—to deliver bioinformatically designed 'mosaic' HIV-1 antigens (Gag, Pol, and Env). These antigens are engineered to provide broad coverage against the global diversity of HIV-1 strains by targeting conserved T-cell and B-cell epitopes. In clinical research, notably studies led by Dr. Boris Juelg, MVA-BN-HIV is evaluated as part of a heterologous prime-boost regimen (typically following Ad26.Mos4.HIV) and often combined with broadly neutralizing antibodies (bNAbs) such as PGT121, PGDM1400, and VRC07-523LS. The goal of these regimens is to induce potent, polyfunctional HIV-specific immune responses to achieve sustained viral remission in the absence of antiretroviral therapy, particularly during analytical treatment interruption (ATI).
