MVA-hliNSmut is a prophylactic vaccine candidate developed as part of a heterologous prime-boost regimen for the prevention of hepatitis C virus (HCV) infection. It utilizes a modified vaccinia Ankara (MVA) viral vector encoding the human MHC class II invariant chain fused to nonstructural antigens from HCV, designed to enhance T cell immune responses. The inclusion of the invariant chain acts as an adjuvant, boosting antigen presentation and increasing both the magnitude and breadth of anti-HCV T cell responses. Preclinical studies demonstrated enhanced proteasomal degradation and immunogenicity, while phase 1 clinical trials showed that MVA-hliNSmut was well tolerated with no serious adverse events reported. The vaccine is being developed primarily for infectious diseases, specifically chronic hepatitis C[1][2][5].
