MVA-NSmut is a recombinant viral vector vaccine candidate designed for the treatment and prevention of Hepatitis C virus (HCV) infection. It utilizes the Modified Vaccinia Ankara (MVA) platform to deliver a genetic sequence encoding the non-structural (NS) proteins of HCV, specifically NS3, NS4, NS5A, and NS5B. The "NSmut" designation indicates that the NS5B polymerase sequence has been genetically modified to abrogate its enzymatic activity, enhancing the safety profile of the vector. MVA-NSmut is typically employed in heterologous prime-boost immunization strategies, often administered as a boost following a priming injection with a simian adenovirus vector such as AdCh3NSmut. This regimen is intended to induce high-frequency, polyfunctional, and multi-specific T-cell responses against conserved HCV antigens to facilitate viral clearance or prevent chronic infection. The asset was originally developed by Okairos (later acquired by GSK) and has been further advanced by ReiThera.
