Overview

MVA RH5 is a viral-vectored vaccine candidate for blood-stage malaria. It consists of a modified vaccinia virus Ankara (MVA) vector genetically engineered to express the _Plasmodium falciparum_ reticulocyte-binding protein homolog 5 (RH5) antigen. The RH5 protein is essential for parasite invasion of red blood cells. By inducing an immune response against RH5, the vaccine aims to prevent red blood cell invasion, stopping blood-stage infection. MVA RH5 is designed to be used as a booster vaccination in a heterologous prime-boost regimen, typically administered 8 weeks after priming with ChAd63 RH5. Clinical trials have shown this regimen induces robust RH5-specific cellular and humoral immune responses, with the MVA RH5 boost significantly enhancing responses compared to the prime alone.
