MVA-Twist-TRICOM (also known as MVA-TWIST/TRICOM) is a recombinant viral vector-based therapeutic cancer vaccine developed by Bavarian Nordic in collaboration with the National Cancer Institute (NCI). The vaccine utilizes a Modified Vaccinia Ankara (MVA) virus vector to deliver the transgene for Twist-related protein 1 (TWIST1), a transcription factor highly expressed in metastatic tumor cells that drives epithelial-to-mesenchymal transition (EMT). Additionally, the vaccine incorporates TRICOM, a triad of T-cell costimulatory molecules consisting of B7-1 (CD80), ICAM-1 (CD54), and LFA-3 (CD58). By expressing both the Twist antigen and costimulatory molecules, MVA-Twist-TRICOM converts infected cells into competent antigen-presenting cells, thereby inducing robust antigen-specific CD4+ and CD8+ T-cell immune responses to target and eliminate metastatic cancer cells.
