**MVA-ZIKV-NS1** is a recombinant viral vector vaccine candidate using a Modified Vaccinia Ankara (MVA) platform to express the non-structural protein 1 (NS1) of Zika virus (ZIKV) from a 2015 Asian isolate (Suriname). It elicits robust humoral and cellular immune responses, including anti-NS1 antibodies and T cell responses, providing 100% protection against lethal intracerebral ZIKV challenge (strain MR766) in immunocompetent mice after a single intramuscular dose. The NS1-targeted approach avoids antibody-dependent enhancement (ADE) risks associated with structural protein vaccines like prM/E, positioning it as a safe, single-dose candidate suitable for vulnerable populations such as immunocompromised individuals, pregnant women, children, and the elderly.[1][5]
