MVA85A is a recombinant viral vector vaccine developed as a candidate booster for tuberculosis (TB) prevention. It consists of the modified vaccinia Ankara (MVA) virus engineered to express antigen 85A, an immunodominant cell-wall protein from Mycobacterium tuberculosis. The vaccine is designed to enhance and prolong the immune response induced by Bacille Calmette–Guérin (BCG), the standard TB vaccine, by boosting antigen-specific T-cell responses, particularly Th1 and Th17 cells. While preclinical studies showed improved protection in animal models when used as a BCG booster, clinical trials in infants and adults—including those with HIV—demonstrated that although MVA85A was safe and highly immunogenic, it did not provide significant additional protection against TB infection or disease compared to BCG alone[1][6][7][8][9].
