MVdeltaC is a novel immunotherapy based on a genetically modified, attenuated Schwarz strain measles virus. It is engineered to selectively replicate in tumor cells that express the CD46 receptor, leading to direct tumor cell lysis and robust activation of the immune system. This dual mechanism aims to induce immunogenic cell death and reactivate "cold" tumors, making them more susceptible to immune attack. Preclinical studies have demonstrated strong anti-tumor activity across multiple human cancer cell lines—including mesothelioma, lung, bladder, ovarian, cervical, and liver cancers—and in animal models. In pleural mesothelioma models specifically, MVdeltaC has shown significant tumor regression and long-term immune protection after treatment. The therapy also produces high levels of defective interfering genomes that may enhance its immune-stimulating effects via the RIG-I pathway. Developed by Oncovita (a spin-off from Institut Pasteur), MVdeltaC received FDA Orphan Drug Designation for pleural mesothelioma in June 2025 and is expected to enter clinical development by 2026[1][2][3][4][5].
