MVF-HER-2 (597-626) + MVF-HER-2 (266-296) + ISA 51 is an investigational combination chimeric B-cell peptide vaccine developed by Pravin T.P. Kaumaya (Ohio State University) and licensed to Imugene. The vaccine consists of two synthetic peptide constructs: MVF-HER-2 (597-626), which mimics the trastuzumab-binding epitope, and MVF-HER-2 (266-296), which mimics the pertuzumab-binding epitope of the HER-2 extracellular domain. Each peptide is fused to a promiscuous T-cell epitope from the measles virus fusion protein (MVF) to enhance immunogenicity and is emulsified in the Montanide ISA 51 adjuvant. The vaccine is designed to stimulate the patient's own immune system to produce a polyclonal antibody response against HER-2, potentially overcoming resistance to monoclonal antibody therapies like trastuzumab and pertuzumab by inducing antibody-dependent cellular cytotoxicity (ADCC) and inhibiting tumor cell signaling. It has been evaluated in Phase I/II clinical trials for advanced solid tumors, including HER-2-positive breast, gastric, and esophageal cancers.
