This is a combination therapeutic cancer vaccine composed of two synthetic chimeric peptides—MVF-HER-2 (597–626) and MVF-HER-2 (266–296)—each representing distinct B-cell epitopes from the human epidermal growth factor receptor 2 (HER-2/neu). These peptides are fused to a measles virus fusion protein T-cell epitope to enhance immunogenicity. The formulation is emulsified in Montanide ISA 720, an experimental water-in-oil adjuvant designed to boost immune responses. The vaccine aims to induce polyclonal antibody responses that mimic those generated by trastuzumab and pertuzumab, targeting both the trastuzumab-binding site in domain IV and the pertuzumab-binding site in domain II of HER‑2. Mechanistically, it stimulates humoral and cellular immunity against HER‑2-positive tumor cells through direct inhibition of HER‑2 signaling pathways as well as antibody-dependent cellular cytotoxicity. It has been evaluated in phase I clinical trials for safety, immunogenicity, and preliminary efficacy in patients with advanced or metastatic solid tumors[1][7][9].
