Overview

MVR-C5252 is a genetically engineered oncolytic herpes simplex virus type 1 (oHSV-1) designed for the treatment of malignant glioma and other high-grade brain tumors. Developed by ImmVira using its OvPENS platform, this recombinant viral therapy is further attenuated from the earlier candidate MVR-T3011 to enhance tumor selectivity and safety. The virus has been modified to express two exogenous genes—interleukin-12 (IL-12), an immunostimulatory cytokine, and an anti-PD-1 monoclonal antibody—to stimulate immune responses within the tumor microenvironment and promote direct oncolytic activity against malignant gliocytes. The drug is currently in Phase 1 clinical trials for recurrent high-grade glioma/glioblastoma in the United States and has received orphan drug designation for glioma[3][4][6][8].
