MVT-1075 is a novel radioimmunotherapy product developed by MabVax Therapeutics in collaboration with Memorial Sloan Kettering Cancer Center. It consists of a fully human monoclonal antibody (HuMab-5B1) that specifically targets the cancer antigen CA19‑9—an antigen overexpressed on pancreatic cancer and other CA19‑9 positive malignancies—conjugated to the beta-emitting radionuclide lutetium‑177 (177Lu). The mechanism of action involves targeted delivery of ionizing radiation to tumor cells expressing CA19‑9 via the antibody-radionuclide conjugate. Preclinical studies demonstrated marked suppression and occasional regression of tumor growth in xenograft models. Phase I clinical trials were initiated for patients with recurrent or metastatic pancreatic ductal adenocarcinoma and other CA19‑9 positive cancers (including colon and lung), primarily to assess safety and dosimetry. The drug showed manageable toxicity at initial doses and evidence of tumor targeting[3][4][8].
