MVT-5873 is a fully human IgG1 monoclonal antibody therapeutic targeting sialyl Lewis A (sLea), an epitope on Carbohydrate Antigen 19-9 (CA 19-9), which is expressed in pancreatic ductal adenocarcinoma and other gastrointestinal and lung cancers. CA 19-9 plays a role in tumor adhesion, metastasis, and is a marker of aggressive tumor phenotype. MVT-5873 binds specifically to sLea antigen on cancer cells, inducing antitumor activity through mechanisms including complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC). It has been developed primarily for treatment of CA 19-9 producing cancers such as pancreatic cancer, cholangiocarcinoma, and metastatic colorectal cancer to the liver. The drug has been studied both as monotherapy and in combination with chemotherapy regimens like gemcitabine plus nab-paclitaxel or modified FOLFIRINOX. Clinical trials have shown that MVT-5873 can reduce serum CA 19-9 levels, decrease tumor volume, prevent tumor progression, and potentially improve recurrence-free survival when used perioperatively around surgical resection.
