MVT-5873 + modified FOLFIRINOX is an experimental combination therapy used in the treatment of CA 19-9–producing cancers, primarily pancreatic, cholangiocarcinoma, and metastatic colorectal cancer to the liver. MVT-5873 is a fully human IgG1 monoclonal antibody that targets carbohydrate antigen 19-9 (CA 19-9), a sialylated glycan highly expressed in several gastrointestinal cancers. MVT-5873 mediates antitumor activity through complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity against tumor cells expressing CA 19-9[1][3][5][7]. Modified FOLFIRINOX is a chemotherapy regimen that typically includes adjustments to the standard FOLFIRINOX dosing/schedule for tolerability and consists of oxaliplatin, irinotecan, leucovorin (folinic acid), and 5-fluorouracil (5-FU)[2][4][6][8]. This combination is being studied in the perioperative setting (around the time of surgery) to assess safety, ability to lower recurrence rates, and potential to extend survival in patients with operable tumors.
